چکیده

A particulate enzyme system from Bacillus sfearofhermophilus which catalyzes the synthesis of peptidoglycan from uridine nucleotide precursors has been investigated. The preparation catalyzed extensive cross-linking of the enzymatically synthesized peptidoglycan. In addition in the presence of ATP and ammonia or L-glutamine amidation of the peptidoglycan also occurred. The nature of the amidated products obtained on lysozyme digestion of the enzymatically synthesized peptidoglycan was elucidated by a comparison with similar materials obtained after lysozyme digestion of isolated vegetative cell walls of this organism.

Synthesis of peptidoglycan by high molecular weight penicillin-binding proteins of Bacillus subtilis and Bacillus stearothermophilus.

The high molecular weight penicillin-binding proteins (PBP(s) ) Bacillus subtilis PBPs 1, 2, and 4 and Bacillus stearothermophilus PBPs 1-4 were shown to catalyze peptidoglycan synthesis from the undecaprenol-containing lipid intermediate substrate in two assay systems. In a filter paper assay system, high levels of substrate polymerization occurred when reaction mixtures were incubated on What...

Reconstitution of Peptidoglycan Cross-Linking Leads to Improved Fluorescent Probes of Cell Wall Synthesis

The peptidoglycan precursor, Lipid II, produced in the model Gram-positive bacterium Bacillus subtilis differs from Lipid II found in Gram-negative bacteria such as Escherichia coli by a single amidation on the peptide side chain. How this difference affects the cross-linking activity of penicillin-binding proteins (PBPs) that assemble peptidoglycan in cells has not been investigated because B....

Kinetics of cross-linking of peptidoglycan in Bacillus megaterium.

The extent and rate of cross-linking of diaminopimelic acid in the peptidoglycan of Bacillus megaterium have been determined by a new procedure. The method is based on the reaction of nitrous acid with the unprotected amino groups of non-cross-linked diaminopimelic acid. Pulse label experiments in a mutant where diaminopimelic acid is incorporated specifically into peptidoglycan demonstrated th...

Structural analysis of Bacillus subtilis spore peptidoglycan during sporulation.

A major structural element of bacterial endospores is a peptidoglycan (PG) wall. This wall is produced between the two opposed membranes surrounding the developing forespore and is composed of two layers. The inner layer is the germ cell wall, which appears to have a structure similar to that of the vegetative cell wall and which serves as the initial cell wall following spore germination. The ...
